Effect of Er'huang Xinshang Zhitong Ointment(二黄新伤止痛软膏)on RIPK1/RIPK3/MLKL-mediated necroptosis in acute soft tissue injury

Abstract
Objective To investigate the effect of Er'huang Xinshang Zhitong Ointment(二黄新伤止痛软膏,EXZO)on RIPK1/RIPK3/MLKL-mediated necroptosis in acute soft tissue injury.Methods Twenty-four 6-8 week-old SPF male SD rats were ran-domly divided into four groups:the normal group(gauze-covered left hindlimb mid-thigh soft tissue fixed with adhesive tape),model group(gauze-covered injured area reinforced with adhesive tape),EXZO treatment group(EXZO applied on injured area reinforced with adhesive tape),and Nec-1(Necroptosis-1)treatment group,with 6 rats in each group.Dressings were changed every 24 hours for 7 consecutive days.24 hours after the last administration,peripheral blood was collected and the damaged tissue part was taken.After the treatment was completed,peripheral blood and damaged tissues were collected.Serum levels of IL-6,IL-1β and TNF-α in ser-um were detected by ELISA.Necroptosis was observed by transmission electron microscopy(TEM),and the protein expressions of caspase-8,MLKL,RIPK1 and RIPK3 were detected by Western blot(WB).Results Compared with the normal group,the contents of IL-6,IL-1β,TNF-α and the protein expressions of MLKL,RIPK1,RIPK3 were increased(P<0.01),while caspase-8 expres-sion was decreased(P<0.01).Compared with the model group,both the EXZO treatment group and the Nec-1 treatment group dem-onstrated a reduction in the contents of IL-6,IL-1β,TNF-α,mitochondrial damage and MLKL/RIPK1/RIPK3 protein expressions(P<0.01),along with increased caspase-8 expression(P<0.01).Conclusion EXZO exerts a positive therapeutic effect on acute soft tissue injury by inhibiting the activation of the RIPK1/RIPK3/MLKL pathway,thereby attenuating necroptosis,and reducing the se-cretion of inflammatory factors.关键词
