石淋清颗粒通过调控SIRT1/PGC-1α信号通路预防大鼠草酸钙肾结石形成的机制研究

Abstract
Objective To investigate the effect of Shilinqing Granule(石淋清颗粒,SLQG)on preventing calcium oxalate kidney stone formation in rats and its influence on the SIRT1/PGC-1α signaling pathway.Methods Fifty SPF SD rats were randomly divided into five groups:the control group,model group,low-dose group,medium-dose group,and high-dose group,with 10 rats in each group.Except for the control group,the calcium oxalate kidney stone model was induced by administering 1%ethylene glycol in drinking water combined with 2%ammonium chloride via gavage for 4 weeks.Simultaneously,the low-doe,medium-dose,and high-dose groups received corresponding doses of SLQG for 4 weeks.After intervention,the following indicators were measured:24-hour urine volume,urinary pH,urinary oxalate(Ox),urinary calcium(Ca2+),serum creatinine(Cr),blood urea nitrogen(BUN),and blood Ca2+.In addition,the renal tissue levels of reactive oxygen species(ROS),malondialdehyde(MDA),and superoxide dismutase(SOD)activity were assessed.Kidney pathological changes were observed using HE staining,and calcium oxalate crystal deposition was examined via Von Kossa staining.Transmission electron microscopy was used to observe renal ultrastructure.Western blot(WB)was conducted to detect the expression of silent information regulator 1(SIRT1)and peroxisome proliferator-activated receptor coactivator-1(PGC-1α)proteins,while PCR was performed to assess SIRT1 and PGC-1α mRNA expression.Results Compared with the model group,the intervention reduced crystalline formation,significantly increased 24-h urine volume and urinary pH in rats(P<0.05),while urinary Ca2+and serum Cr and BUN levels were significantly reduced(P<0.05);meanwhile,ROS and MDA levels were reduced and SOD activity was increased(P<0.05);mitochondrial damage in peduncle cells and renal tubular cells was significantly mitigated,and SIRT1 and PGC-1α were highly expressed(P<0.05).Conclusion SLQG can prevent calcium oxalate kidney stone formation in rats,reduce ROS production,and mitigate mitochondrial damage in podocytes and the renal tubular epithelial cells.These effects may be associated with the activation of the SIRT1/PGC-1α signaling pathway.关键词
