Production of frozen semen and test of artificial insemination in Xizang Niangya yak

Abstract
In order to explore the efficient propagation technique of Niangya yak,the semen of 3 Nyonya male yak was collected using the false vagina method,and the quality of the sperms was detected,and thin tube frozen semen was prepared.Then,fifty-six non-pregnant healthy female yak aged about 3 years old were selected for simultaneous estrus treatment with three drug combinations PG+PG+PMSG(Group A,n=16),Cue-Mate+PG+PMSG(Group B,n=20)and GnRH+PG+GnRH(Group C,n=20),and artificial insemination was performed.Finally,pregnancy detection was carried out with early pregnancy test card and portable B-ultrasound diagnostic instrument.The results showed that the average semen volume,sperm density,viability and deformability in Niangya yak were(2.03±0.25)mL,(14.04±2.52)×108/mL,(83.00±3.46)%and(5.33±1.53)%,respectively;which met the preparation criteria of frozen semen.After being thawed,the average sperm motility rate was(46.08±2.14)%,with a malformation rate of(10.72±5.81)%,a plasma membrane integrity rate of(47.69±1.42)%,and an acrosome integrity rate of(55.94±1.75)%.The results also showed that the estrus rate of Group B was 60%at 48 h,that of Group A was 37.5%at 48 h,that of Group C was the slowest,and that of Croup C was only 20%at 48 h and 60%at 96 h.The highest total estrus rate was 90%in Group B,which was not significantly different from 87.5%in Group A(P>0.05),but signif-icantly different from 60%in Group C(P<0.05).The pregnancy rates of the three groups were 28.57%,33.33%and 33.33%,respective-ly,with no significant difference(P>0.05).This paper provided practical guidance for the preparation and artificial insemination of fine tube frozen essence in Niangya yak.关键词
