Abstract
In September 2024,an outbreak suspected of Clostridium perfringens(C.perfringens)infection occurred in a goat farm in Gaotang County,Shandong Province.In order to confirm the pathogen serotypes and their genetic evolutionary characteristics to take specific measures,livers,lungs,spleens and other tissue samples of sick goats were collected for bacterial isolation and culture,then PCR amplification and sequencing of 16S rDNA were conducted based on the isolates,a phylogenetic tree was drawn using MEGA 7.0 software,and 7 pairs of toxin genotyping primers of C.perfringens were used to identify the serotypes,followed by the mouse pathogenicity test.The results showed that 5 strains consistent with the characteristics of C.perfringens were isolated,16S rDNA genes of which were amplified by PCR to obtain bands with 1 400 bp,with 99.6%above similarity to the reference strain of C.perfringens,locating in the same sub-branch with Shanghai human-derived 218002-301 and Indian leopard-derived ATCC 13124 strain,indicating a close genetic relationship,but far away from Tibet yak-derived BG01,East China rabbit-derived SD14-02,Yunnan dairy goat-derived KM-1,South Korea donkey-derived KSJ-28 and Beijing human-derived 1005-15098 strain;a band of 402 bp was obtained after PCR amplification of toxin genes,conforming to the size of α-toxin gene,and the obtained sequence was 99.9%similar to that of C.perfringens type A,therefore confirmed as C.perfringens type A.Mouse pathogenicity test showed that the isolates were highly pathogenic.In conclusion,C.perfringens type A was responsible for the death of the goats.Local farms were recommended to strengthen their feeding management,regularly disinfect animal houses and environment,and to formulate a reasonable immunization plan to prevent any occurrence or spread of C.perfringens.Some reference was provided for future diagnosis and treatment of C.perfringens infection.关键词
