6种猪病毒病微流控芯片多重检测方法的建立OA
Establishment of a Microfluidic Chip Method for Multiple Detection of 6 Porcine Viral Diseases
为实现对猪繁殖与呼吸综合征病毒(PRRSV)经典毒株、PRRSV高致病性毒株(HP-PRRSV)、猪瘟病毒(CSFV)、猪伪狂犬病病毒(PRV)、猪圆环病毒 2 型(PCV2)和猪细小病毒(PPV)的多重检测,根据此 6 种病毒的基因保守区域分别设计引物,将引物包埋在恒温扩增微流控芯片的反应池内,通过优化引物浓度、反应温度和反应时间,建立了可同时检测上述 6 种病毒的碟式微流控芯片方法.结果显示:建立的微流控芯片方法对 6 种病毒质粒模板的最低检测限为 103 copies/µL;特异性分析显示,病毒或样品只在与其相对应检测指标的反应池内有"S"形扩增曲线;重复性良好,批内和批间变异系数均小于 5%;该方法与商品化荧光定量检测试剂盒对 40 份临床样品的检测符合率达到 95%以上.结果表明,本研究成功建立了 6 种猪病毒病微流控芯片检测方法,其灵敏度高、特异性强、重复性好,实现了对多种动物疫病的联合检测.
In order to simultaneously detect the classical strain of porcine reproductive and respiratory syndrome virus(PRRSV),PRRSV highly pathogenic strain(HP-PRRSV),classical swine fever virus(CSFV),pseudorabies virus(PRV),porcine circovirus type 2(PCV2)and porcine parvovirus(PPV),primers were designed according to the genetically conserved regions of the 6 viruses,respectively,and embedded in reaction pools of isothermal amplification microfluidic chip,a microfluidic chip method was established to simultaneously detect all the above viruses through optimizing the concentration of primers,reaction temperature and time.The results showed that the lowest detection limits of the established method for the plasmid templates of the 6 viruses were all 103 copies/µL;specificity analysis showed that viruses or samples presented an"S"-shaped amplification curve only in the reaction pool corresponding to their specific detection indicators;good reproducibility was observed,the intra-and inter-batch coefficients of variation were less than 5%;for 40 clinical samples,the compliance rate of the method with commercial quantitative fluorescence detection kits reached above 95%.In conclusion,a microfluidic chip method was successfully established for 6 porcine viral diseases,which presented high sensitivity,strong specificity and good reproducibility,achieving joint detection for multiple animal diseases.
