非洲猪瘟病毒抗体阻断ELISA法测量不确定度评估OA

为了评估阻断ELISA方法检测结果的可靠性,使用阻断ELISA方法检测非洲猪瘟病毒(ASFV)抗体,并分别使用"影响因素分析"法和"对照样品"法对试验进行测量不确定度评估."影响因素分析"法和"对照样品"法评估的扩展不确定度(U)分别为 0.053 0 和 0.044 2(k=2),表明在控制好移液器、培养箱和酶标仪等仪器引入的不确定度基础上,ELISA方法检测ASFV抗体的整体不确定度相对较低.两种方法评估的不确定度值差异不显著,使用"影响因素分析"法可以通过分析各分量在测量结果中的相对贡献,得到影响结果的关键因素,并进行改进;使用"对照样品"法可减少大量计算过程,操作更简单.ASFV抗体ELISA试验判定引入不确定度,可以提高结果的准确性,有利于进一步完善兽医检测领域不确定度评估体系.

In this study,blocking ELISA was used to detect antibodies against African swine fever virus(ASFV)for the purpose of evaluation on its reliability,of which measurement uncertainty was assessed by the"influence factor analysis"and the"control sample"method,respectively.Results showed that the expanded uncertainty values were determined as 0.053 0 and 0.044 2(k=2)by the two methods,indicating an overall low uncertainty of ELISA for ASFV antibodies on the basis of controlling the uncertainty that might be caused by the pipettes,incubators or microplate reader.No significant difference was observed between the uncertainty values evaluated by the two methods.Key factors influencing the results were obtained by the"influence factor analysis"method through analyzing relative contribution of each component to the results,thus supporting further improvement;while a large number of calculations might be reduced by the"control sample"method,which was more accessible.In conclusion,accurate results could be obtained by the application of uncertainty in ELISA assay for ASFV antibodies,contributing to the improvement of uncertainty evaluation system in veterinary testing.
