Mechanism of lncRNA SERPINB9P1/miR-655-3p/CXCR4 pathway in the transdifferentiation of prostatic fibroblasts to myofibroblasts

Abstract
[Objective]To explore the role of lncRNA SERPINB9P1 in the transdifferentiation of prostatic fibroblasts(FB)to myofibroblasts(MFB)and analyze its regulatory mechanisms.[Methods]An FB to MFB transdifferentiation model was established using CXCL12.The expression of lncRNA SERPINB9P1 and miR-655-3p was detected by qPCR,and the expression of α-SMA,COL1,and CXCR4 was assessed by qPCR and Western blot.Bioinformatics tools were used to predict the binding sites between miR-655-3p,lncRNA SERPINB9P1,and CXCR4.Dual-luciferase reporter assays were performed to examine the interaction between miR-655-3p,lncRNA SERPINB9P1,and CXCR4.CCK-8 assays were used to assess cell proliferation.[Results]The FB-MFB cell model exhibited significant morphological changes,enhanced proliferation,and upregulated expression of transdifferentiation markers α-SMA and COL1.Moreover,the expression of lncRNA SERPINB9P1 was significantly increased in the FB-MFB model.Knockdown of lncRNA SERPINB9P1 inhibited the expression of α-SMA and COL1 and significantly suppressed cell proliferation,while overexpression of lncRNA SERPINB9P1 promoted the expression of these markers and enhanced cell proliferation.Further studies showed that lncRNA SERPINB9P1 could mediate the expression of CXCR4 to promote FB to MFB transdifferentiation.Bioinformatics prediction and experimental validation revealed that miR-655-3p interacted with lncRNA SERPINB9P1 and CXCR4.miR-655-3p mimic treatment suppressed CXCR4 expression,while overexpression of lncRNA SERPINB9P1 indirectly upregulated CXCR4 expression by inhibiting miR-655-3p.[Conclusion]LncRNA SERPINB9P1 promotes FB to MFB transdifferentiation by inhibiting miR-655-3p expression and thereby upregulating CXCR4 expression.关键词
