清热化湿通络方对慢性萎缩性胃炎癌前病变模型大鼠的保护作用及其作用机制
Protective Effect and Mechanism of Qingre Huashi Tongluo Formula on Chronic Atrophic Gastritis Precancerous Lesion Model Rats

Abstract
Objective To investigate the protective effect and mechanism of Qingre Huashi Tongluo Formula on chronic atrophic gastritis(CAG)precancerous lesion model rats.Methods Eight rats were randomly selected for normal feeding and set as the blank control group(group A).The other 46 rats were used to replicate the CAG precancerous lesion rat model by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)complex method,and the 40 successfully modeled rats were randomly divided into the model control group(group B),the positive drug group(group C,400 mg/kg Weifuchun Capsules),and low-,medium-,and high-dose groups of Qingre Huashi Tongluo Formula(groups D1,D2,D3,100,200,400 mg/kg),with eight rats in each group.Rats in groups C,D1,D2,and D3 were orally administered with corresponding drugs,while rats in groups A and B were orally administered with 10 mL of 0.1%sodium chloride solution for 12 weeks.Hematoxylin-eosin staining method was used to analyze pathological tissue damage and evaluate the histological damage index(HDI).The fully automated biochemical analyzer was used to detect the levels of gastric function-related indicators.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of inflammatory factors.Western blot was used to detect the expression levels of signaling pathway-related proteins.Results Compared with those in group B,the atrophy morphology of gastric tissue and pathological damage of gastric mucosa in groups C,D2,and D3 improved;the levels of serum pepsinogen Ⅰ(PG Ⅰ),pepsinogen Ⅱ(PG Ⅱ),and gastrin(GAS)in groups C,D2,and D3 significantly increased(P<0.05);the HDI score and levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),prostaglandin E2(PGE2),and the expression levels of phosphorylated p-65(p-p65)/p65,phosphorylated Kappa B inhibitory protein(p-IκB)/IκB,phosphorylated phosphatidylinositol 3-kinase(p-PI3K)/PI3K,and phosphorylated serine-threonine protein kinase(p-Akt)/Akt proteins in gastric mucosal tissue in groups C,D2,and D3 significantly decreased(P<0.05).Conclusion Qingre Huashi Tongluo Formula can alleviate gastric mucosal damage and inflammatory response in CAG precancerous lesion model rats,improve gastric function,and may be achieved by inhibiting the activation of the PI3K/Akt/nuclear factor(NF-κB)signaling pathway.关键词
