基于主成分分析法探讨红芪配伍淫羊藿治疗骨质疏松症模型大鼠的疗效研究
Therapeutic Effect of Hedysari Radix Combined with Epimedii Folium on Osteoporosis Model Rats Based on Principal Component Analysis

Abstract
Objective To investigate the synergistic therapeutic effect of Hedysari Radix combined with Epimedii Folium on osteoporosis(OP)in ovariectomized rats.Methods A total of 48 female SD rats were randomly divided into the blank control group(group A,equal volume of physiological saline),the model group(group B,equal volume of physiological saline),the positive drug control group(group C,0.2 g/kg Xianling Gubao Capsules),the Hedysari Radix-Epimedii Folium group[group D,10 g/kg Hedysari Radix-Epimedii Folium solution(calculated by the crude herbs)],the Epimedii Folium group[group E,10 g/kg Epimedii Folium solution(calculated by the crude herbs)],and the Hedysari Radix group(group F,10 g/kg Hedysari Radix solution(calculated by the crude herbs)],with eight rats in each group,and the OP models were replicated.After successful modeling,rats in each group were orally administered with corresponding drugs for three consecutive months.The bone mineral density(BMD)of rats'whole body and BMD of rats'right femur,the related bone mechanics indicators(maximum load capacity and elastic modulus),biochemical indicators[serum calcium(Ca),phosphorus(P),alkaline phosphatase(ALP),and tartrate resistant acid phosphatase(TRACP)]were measured.The principal component analysis(PCA)method was used to weight and sum the OP-related indicators of rats in each group,and the comprehensive score ranking was obtained.The organ coefficient was measured and the pathological changes of femur tissue were observed by the Hematoxylin-eosin(HE)staining method.Results Compared with those in group B,the BMD,maximum load value,elastic modulus,serum calcium,and phosphorus in group D significantly increased(P<0.05),while the TRACP in group D significantly decreased(P<0.05).The PCA results showed that group D had the highest comprehensive score in the treatment group,indicating that the combination of Hedysari Radix and Epimedii Folium had the best effect in the treatment of OP.Compared with those in group B,the coefficients of liver,heart,lungs,kidneys,and spleen in the other groups had no significant difference(P>0.05),but the uterine coefficient in the other groups significantly increased(P<0.05).The bone trabeculae in group D increased and widened,with an increase in osteoblasts and a significant increase in hematopoietic cells.Conclusion Hedysari Radix combined with Epimedii Folium has a good therapeutic effect and safety in the treatment of OP model rats,which can improve BMD,bone mechanics indicators,biochemical indicators,increase the number of osteoblasts,and reduce trabecular degeneration and necrosis.关键词
