基于指纹图谱和含量测定的蜜远志炮制研究
Processing of Polygalae Radix Praeparata Cum Melle Based on Fingerprints and Content Determination

Abstract
Objective To establish an ultra-high performance liquid chromatography(UPLC)fingerprints and content determination method for Polygalae Radix Praeparata Cum Melle,and to investigate the change rule in chemical components of Polygalae Radix Praeparata Cum Melle before and after processing.Methods The UPLC fingerprint of Polygalae Radix Praeparata Cum Melle was established,the common peaks were identified,and the similarity was evaluated by the Similarity Evaluation System for Chromatographic Fingerprinting of Traditional Chinese Medicine(2012 Version),and characteristic peaks were identified through high-resolution mass spectrometry.Combining chemometric analysis,partial least squares-discriminant analysis(PLS-DA)was used for clustering analysis of chemical components;the content determination of tenuifolin,polygalaxanthone Ⅲ and 3,6'-disinapoyl sucrose were determined by the UPLC method.Results Nine common peaks were determined in the fingerprint of Polygalae Radix Praeparata Cum Melle,namely sibiricose A5,sibiricose A6,polygalaxanthone Ⅲ,3,6'-disinapoyl sucrose,arillanin A,tenuifolin A,tenuifoliose H,tenuifoliose A and 1,2,3,7-tetramethoxyxanthone.PLS-DA could classify Polygalae Radix and Polygalae Radix Praeparata Cum Melle into two categories and the main differential components were peak 8(tenuifoliose A)and peak 9(1,2,3,7-tetramethoxyxanthone).The content determination results showed that the contents of tenuifolin,3,6'-diferuloyl sucrose,and polygalaxanthone Ⅲ in Polygalae Radix before processing were 21.06,6.09,and 0.97 mg/g,which were significantly higher than 17.63,5.47,and 0.78 mg/g after processing(P<0.05).Conclusion The established UPLC fingerprints and content determination method for Polygalae Radix Praeparata Cum Melle are accurate,efficient,and highly specific,which can provide a reference for the optimization of processing technology and quality evaluation of Polygalae Radix Praeparata Cum Melle.关键词
