Effects of EA Intervention on Ferroptosis-Related Protein Expressions in Chondrocytes of KOA Rabbits

Abstract
Objective:To observe the effect of electro-acupuncture(EA)intervention on the ferroptosis-related protein expressions of SLC7A11 and GPX 4,as well as on the level of GSH in rats with knee osteoarthritis(KOA).Methods:35 rabbits were randomly divided into the normal group,the model group,the model inhibitor group,the EA group,and the EA inhibitor group,with 7 rats in each group.The KOA model was eatablished using the modified Videman immobilization method with the left hind limb in an extended position.The normal group and the model group received no intervention,whereas the EA group and the EA hihibitor group were treated for 3 weeks.Transmission electron microscopy was used to observe the morphological changes in chondrocytes and mitochondria.Western blot method was performed to detect the protein expressions of SLC7A11 and GPX 4 in the knee joint cartilage.Biochemical method was used to measure the GSH content in the knee joint cartilage.Results:Compared with the normal group,the model group exhibited chondrocyte membrane rupture,organelle swelling,increased mitochondrial membrane density,reduced or absent cristae,and outer membrane ruptured;the protein expressions of SLC7A11 and GPX 4 were significantly decreased(P<0.01),and the level of GSH was also significantly reduced in the model group compared to those in the normal group(P<0.01).Compared with the model group,the EA group exhibited normal chondrocyte and mitochondrial morphology,with increased levels of SLC7A11,GPX 4 and GSH(P<0.01).Compared with their respective intervention groups,the model inhibitor group and the EA hihibitor group showed decreased levels of SLC7A11,GPX 4 and GSH(P<0.01,P<0.05).Conclusion:EA intervention can improve the pathological state of chondrocyte ferroptosis,which may be related to up-regualting the ferroptosis-related proteins expressions of SLC7A11,GPX 4 and GSH.关键词
