基于网络药理学和分子对接技术探讨贯叶金丝桃治疗卒中后抑郁作用机制
Exploration on the mechanism of Hypericum perforatum in the treatment of post-stroke depression based on network pharmacology and molecular docking

Abstract
Objective:To explore the action mechanism of Hypericum perforatum in the treatment of post-stroke depression based on network pharmacology and molecular docking.Methods:The active ingredients and target of Hypericum perforatum were obtained by TCMSP,and the target was supplemented by Swisstarget database and TargetNet database.OMIM database and Genecards database were used to obtain the related targets of post-stroke depression.The intersection targets of Hypericum perforatum and post-stroke depression were obtained by Venny 2.1.0 platform.The network of"TCM-Active Ingredient-Target"was constructed using Cytoscape 3.7.2.Based on the data from STRING platform,the protein-protein interaction network was constructed on the intersection targets by Cytoscape software,and the core targets were screened.The KEGG pathway and GO function enrichment analysis were carried out through DAVID database,and the results were visualized by R version 3.7.2 software.Finally,Autodock vina software was used to study the molecular docking.Results:A total of 7 active ingredients and 177 targets of Hypericum perforatum were obtained,and 5516 targets related to post-stroke depression were obtained.After intersection,153 potential targets of Hypericum perforatum on post-stroke depression were obtained,and the targets with high correlation were AKT1,PTGS2,TNF,JUN and MAPK8.It was mainly enriched in PI3K-Akt signaling pathway,AGE-RAGE signaling pathway in diabetic complications,TNF signaling pathway,IL-17 signaling pathway,chemical carcinogenesis-reactive oxygen species,chemical carcinogenesis-receptor activation,fluid shear stress and atherosclerosis.Conclusion:Hypericum perforatum may regulate apoptosis and differentiation of cells,inhibit inflammatory response,improve antioxidant capacity and so on,so as to treat post-stroke depression.关键词
