基于数据挖掘与网络药理学探讨内服中药复方治疗风瘙痒(血虚风燥证)的用药规律及机制
Exploring the medication rule and mechanism of oral compound formulas of Chinese medicine in treating pruritus cutanea of syndrome of wind-dryness due to blood deficiency based on data mining and network pharmacology

Abstract
Objective:Exploring the medication rule of oral compound formulas of Chinese medicine for the treatment of pruritus cutanea of syndrome of wind-dryness due to blood deficiency based on the China Patent Authorization Announcement,CNKI,VIP,and Wangfang data,and analyzing the mechanism of action of the top 10 most frequently used Chinese medicines.Methods:We searched the official website of China Patent Authorization Announcement,CNKI,VIP,and Wangfang data to obtain the original data of compound formulas of Chinese medicine for the treatment of pruritus cutanea of syndrome of wind-dryness due to blood deficiency,and then used Excel to perform frequency,medicinal nature and flavour,channel tropism statistics on the prescriptions,and then used IBM SPSS Modeler and IBM SPSS Statistics to perform the association rule analysis and clustering analysis.The medication rule of compound formulas of Chinese medicine for the treatment of pruritus cutanea of syndrome of wind-dryness due to blood deficiency was analyzed.Based on the network pharmacology method,we predicted the main components,targets of the drugs and disease pathways,constructed the"drug-component-target-pathway-disease"diagram network,obtained the disease targets,gained the drug-disease intersection targets through the Venny 2.1.0 online platform,and then performed the intersection targets GO and KEGG enrichment analyses.Finally,AutoDock 15.6 was utilized to perform molecular docking of the main components and targets.Results:A total of 142 compound formulas of Chinese medicine and 216 Chinese medicines were included in the data mining,among which 21 high-frequency Chinese medicines were included.A total of 158 association rules were obtained,and cluster analysis revealed that there were 7 clusters when the pitch was 25.The top 10 most freguently used Chinese medicines were selected for network pharmacology analysis,including Danggui(Radix Angelicae Sinensis),Baishao(Radix Paeoniae Alba),Shengdihuang(Radix Rehmanniae),Fangfeng(Radix Saposhnikoviae),Gancao(Radix Glycyrrhizae),Jili(Fructus Tribuli),Baixianpi(Cortex Dictamni),Jingjie(Herba Schizonepetae),Shudihuang(Radix Rehmanniae Preparata),Chuanxiong(Rhizoma Chuanxiong).There were 99 core active ingredients and 249 action targets,and the molecular docking results suggested that the core ingredients were stably bound to the core targets,with the lowest binding energy of-8.66 kcal/mol and the average binding energy of-6.23 kcal/mol.Conclusion:Most of the medicines used in the treatment of pruritus cutanea of syndrome of wind-dryness due to blood deficiency with compound formulas of Chinese medicine were based on the methods of tonifying,dispelling wind,and relieving the epidemiology,and were supplemented with blood invigorating,heat clearing,and heat removing medicines.The core drugs mainly act on targets such as IL-6,AKT1,and EGFR,and signaling pathways such as lipid and atherosclerosis,and prostate cancer,thus providing a reference for the clinical use of compound formulas of Chinese medicine in the treatment of pruritus cutanea of syndrome of wind-dryness due to blood deficiency.关键词
