Exploring the causal relationship between circulating inflammatory proteins and colorectal cancer through two-sample Mendelian randomization study

Abstract
Objective:To investigate the causal relationship between circulating inflammatory proteins and colorectal cancer.Methods:The instrumental variable data for 91 circulating inflammatory proteins was derived from a genome-wide association study(GWAS)involving 14 824 participants with a predominantly European population.The GWAS data for colorectal cancer came from the Finnish database(R10),including 6 847 cases and 314 193 controls.Mendelian randomization analysis was conducted using methods such as inverse-variance weighting(IVW),weighted median,MR-Egger regression,and weighted mode.Results:According to the IVW analysis,Delta and Notch-like epidermal growth factor receptor(DNER)level and vascular endothelial growth factor A(VEGFA)level were associated with an increased risk of colorectal cancer(OR=1.122,95%CI:1.012-1.244,P=0.029;OR=1.087,95%CI:1.015-1.165,P=0.017).Conversely,increased axis inhibition protein-1(Axin-1)level and Fms-related tyrosine kinase 3 ligand(FLT3L)level were associated with a reduced risk of colorectal cancer(OR=0.841,95%CI:0.714-0.991,P=0.038;OR=0.917,95%CI:0.847-0.993,P=0.034).Sensitivity analysis further excluded the impact of heterogeneity and pleiotropy.Conclusion:This study identified four circulating inflammatory proteins causally associated with colorectal cancer.Elevated levels of DNER and VEGFA are risk factors for colorectal cancer,while increased levels of Axin-1 and FLT3LG serve as protective factors.关键词
