基于网络药理学和分子对接探讨柴芩温胆汤治疗睡眠障碍的作用机制
The action mechanism of the Chaiqin Wendan decoction on sleep disorder based on network pharmacology and molecular docking

Abstract
Objective:To investigate the action mechanism of the Chaiqin Wendan decoction(柴温胆汤)in the treatment of sleep disorder by network pharmacology and molecular docking.Methods:The chemical constituents and targets of the Chaiqin Wendan decoction,and sleep disorder targets were searched in TCMSP,Pubchem database,SwissADME platform and GeneCards database to obtain the intersected target genes of the Chaiqin Wendan decoction and sleep disorder.A protein-protein interaction network was constructed through STRING database and Cytoscape software.The intersecting targets were imported into DAVID database and R language software for GO function and KEGG pathway enrichment analyses.The core targets were imported into Cytoscape 3.10.0 software to draw the drug-active ingredient-common targets-disease network map,and finally AutoDuck was used to verify the molecular docking between the key active ingredients of the Chaiqin Wendan decoction and the core targets.Results:Through screening,a total of 189 active ingredients and 1017 targets were obtained from the Chaiqin Wendan decoction,and 3 565 disease targets of sleep disorder and 557 intersecting targets of the two were gained.β-sitosterol,baicalein,isorhamnetin,kaempferol,and quercetin were the main active ingredients of the Chaiqin Wendan decoction.The therapeutic targets of sleep disorder were glyceraldehyde-3-phosphate dehydrogenase,serine/threonine protein kinase 1,interleukin-6,tumor necrosis factor,non-receptor tyrosine kinase SRC proto-oncogene,etc.The main signaling pathways involved in regulation included AGE-RAGE signaling pathway in diabetic complications,PI3K-Akt signaling pathway,lipid and atherosclerosis,chemical carcinogenesis-receptor activation signaling pathway,etc.Conclusion:This study predicts the active ingredients,action targets and regulatory pathways of the Chaiqin Wendan decoction in the treatment of sleep disorder,providing a theoretical basis for subsequent studies.关键词
