摘要

Abstract

Objective To investigate the antibacterial activity of cinnamaldehyde against Mycobacterium tubercu-losis(MTB),and to further understand the intracellular bactericidal effect of cinnamaldehyde and its mecha-nism.Methods Determination of the minimum inhibitory concentration(MIC)and the minimum bactericidal concentration(MBC)of cinnamaldehyde against Mycobacterium tuberculosis standard strain H37Rv by applying microdilution resazurin titration;The intracellular bactericidal effect of cinnamaldehyde was assessed by counting intracellular viable colonies;Lactic dehydrogenase(LDH)assay was used to detect the cytotoxic effect of cinna-maldehyde;Flow cytometry was used to detect the apoptosis of macrophages after H37Rv infection treated with cinnamaldehyde,and Western blot(WB)was used to detect the changes of apoptosis and autophagy of macro-phages after cinnamaldehyde treatment.Results Cinnamaldehyde had a strong extracellular antibacterial and bac-tericidal activity against H37Rv with MIC of 59 μmol/L and MBC of 59-118 μmol/L;Cinnamaldehyde has an intracellular bactericidal activity against H37Rv infected macrophages,and has no obvious cytotoxicity at the minimum inhibitory concentration;Flow cytometry and WB showed that cinnamaldehyde promoted the apoptosis of macrophages after H37Rv infection.Conclusion Cinnamaldehyde has bactericidal activity against Mycobacteri-um tuberculosis,and may play an intracellular role by inducing macrophage apoptosis.
