摘要

Abstract

[Objective]Agrobacterium-mediated genetic transformation of soybean cotyledonary nodes is a key technology to achieve targeted improvement of soybean varieties,but it is susceptible to the limitations of the genetic background,and the receptor genotype is one of the key factors affecting the transformation efficiency.Therefore,this study aimed to establish a genetic transformation system for major cultivated varieties,so as to provide a foundation for achieving targeted improvement of soybean cultivars.[Method]In this research,the plant expression vector containing GUS reporter gene was used to optimize the agrobacterium-mediated genetic trans-formation system of soybean cotyledon nodes.[Result]During the co-culture period,positioning explants hori-zontally with the adaxial surface upward and supplementing the medium with 150 mg·L-1 dithiothreitol(DTT)reduced tissue browning by 15%and increased bud induction rate by approximately 15%.Application of this op-timized system to 14 elite soybean cultivars revealed distinct genotypic variations.High regeneration and trans-formation efficiencies were observed in Zhonghuang 13,Huachun 6,and Jidou 17 with the highest value of 5.30%,10.22%and 4.20%,respectively,while Heihe45 exhibited strong regeneration capacity but low transformation efficiency with the maximum transformation efficiency of 2%.Cultivars including Hefeng 55,Hefeng 50,Henong 71,Mengdou 1137,Heike 60,Henong 85,Heike 88,Henong 95,Heinong 84,and Heinong 531 demonstrated both low regeneration and transformation efficiencies(less than 2%).[Conclusion]The findings establish a foundation for achieving targeted improvement of major soybean cultivars.
