摘要

Abstract

[Objective]This research aimed to provide a molecular bioinformatics basis for constructing vec-tors and functionally validating VcGA2ox2 in blueberry.[Method]Using exogenous GA3 to treat fertilized blue-berry flowers and obtain seedless mature blueberry fruits as the materials,a differentially expressed gene named VcGA2ox2 was screened using transcriptome data,and gene cloning and bioinformatics analysis were performed on it.[Result]VcGA2ox2 gene was successfully cloned and analyzed,containing an open reading frame(ORF)of 996 bp,encoding 331 amino acids with two conserved domains(2OG-FeII-Oxy and DIOX-N),belonging to a hydrophilic protein.The phylogenetic tree analysis of amino acid sequences showed that the VcGA2ox2 protein was closely related to CsGA2ox2 and CrGA2ox2 in the Camelliaceae family;qRT-PCR analysis results showed that VcGA2ox2 was expressed in blueberry flowers,leaves,stems,fruits and roots,and it had tissue specificity,with the highest expression level in the roots.GA3 treatment could up-regulated the expression of VcGA2ox2 in fruits at the two stages.[Conclusion]It was speculated that GA3 treatment can induce high expression of GA2ox2,promote GA inactivation in blueberry fruits,and inhibit ovule development,resulting in seedless blueberry fruits.
