Abstract

[Objective]This study aimed to increase leaf yield and provide a reference for improving leaf quality of Ginkgo biloba leaf-using forest by pre-treating the plantations with salicylic acid(SA)and methyl jasmonate(MJ)prior to leaf harvesting,to explore the physiological mechanisms by which SA and MJ regulate leaf growth and flavonoid metabolism in ginkgo.[Method]One month before harvesting G.biloba leaves in autumn,seedlings of approximately three years old were treated with 0,50,100 μmol·L-1 SA,100,250 μmol·L-1 MJ and 100 μmol·L-1 SA+250 μmol·L-1 MJ,respectively.The effects of preharvest SA and MJ treatments on leaf yield,photosynthetic physiology,key enzymes of flavonoid metabolism and flavonoid yield in G.biloba were investigated.[Result]The treatment with 100 μmol·L-1 SA or MJ increased the content of photosynthetic pigments in the leaves,enhanced the photosynthetic efficiency by increasing the maximum photochemical efficiency(Fv/Fm),effective photochemical efficiency(Fv'/Fm')and electron transport efficiency(ETR).For example,the level of Fv'/Fm'increased by 74.0%under SA2 treatment compared with the control,however,no significant effect on photosynthetic pigments was found in the MJ2(250 μmol·L-1)treatment;the values of photochemical quenching coefficient(qP)and heat dissipation coefficient(D)in the MJ2 treatment were higher than those in the SA2 and MJ1 treatments,which indicated the higher concentration of MJ(250 μmol·L-1)could induce mild abiotic stress on the leaves.SA1 or SA2 treatments were beneficial for increasing cinnamyl alcohol dehydrogenase(CAD)or peroxidase activity,respectively,while MJ2 treatment was more beneficial for enhancing phenylalanine ammonlyase(PAL)activity.The 100 and 250 μmol·L-1 MJ treatments significantly in-creased the leaf fresh yield per plant(LYP)and the total flavonoids yield per plant(FYP).The LYP was 96.0%and 137.0%,and FYP was 145.2%and 258.0%higher in 100 or 250 μmol·L-1 MJ treatment than that in the con-trol,respectively.Both the SA treatment alone and the mixed SA+MJ treatment increased the LYP and FYP values of the Ginkgo leaf-using forest,although the effect was not as good as that in the MJ treatment.[Conclusion]Both SA and MJ could promote the growth of preharvest G.biloba leaves in autumn,and increase the yield of leaves and total flavonoids.Treatment with 100 and 250 μmol·L-1 MJ had the best effect.This study provides a strong theoretical basis and technical support for the cultivation and efficient production of G.biloba leaf-using forests.
