摘要

Abstract

[Objective]The relationship between PbCYP86A1,PbCYP86B1 genes and pear peel brown traits was explored.[Method]The PbC-YP86A1/B1 gene was cloned from the peel of"Dangshansu"pear and"Dangshanjinsu"pear by reverse transcription polymerase chain reac-tion(RT-PCR).The virus-induced gene silencing(VIGS)method was used to silence the target gene using the tobacco crispy crack virus(TRV)vector.By analyzing the control group(TRV2-TRV1)and the experimental group(TRV-PbCYP86A1,TRV-PbCYP86B1)pear peri-carp phenotype,the chromatic aberration,relative expression of the gene and changes in the composition of the suberin.[Result]The results showed that after TRV-PbCYP86A1 and TRV-PbCYP86B1 were injected into the pear epidermis,suberin in the injection parts could not be accumulated with the fruit development and exhibited with yellow-green,the color difference between the injection site and the surrounding per-icarp was significantly greater than that of the control group.qRT-PCR indicated that the expressions of PbCYP86A1 and PbCYP86B1 genes re-duced significantly with significant difference in the exocarp of'Xiusu'pear with VIGS compared with control.[Conclusion]The results of su-berin content analysis showed that both PbCYP86A1 and PbCYP86B1 gene silencing could significantly inhibit the synthesis of C16ω-hydroxy-fatty acids and C16 α,ω-diacid in pear pericarp,which will reduce the total amount of suberin.
