摘要

Abstract

Using sea-buckthorn branches as raw materials,the extraction process of total flavonoids was optimized using single factor experi-ments and response surface methodology,and its protective effect on alcoholic liver injury(ALI)was studied.The results showed that the total flavonoid content in the branches and trunks of sea-buckthorn was 0.64%.The optimal extraction process for total flavonoids from sea-buck-thorn branches was ultrasonic frequency of 50 kHz,power of 200 W,extraction temperature of 55℃,liquid-material ratio of 23:1,ethanol con-centration of 61%,extraction time of 2 hours.Under these conditions,the total flavonoid extraction amount was 4.19 mg/g.Total flavonoids were purified and enriched using X-5 macroporous resin.In vitro antioxidant tests showed that the 40%ethanol purified product had the best antioxidant activity.The scavenging rates of DPPH,ABTS,hydroxyl and superoxide anion radicals were 3.17,37.06,792.51 and 262.42 μg/mL,respectively.Cell experiments showed that total flavonoids from sea buckthorn branches could effectively reduce the damage of ethanol to HepG2 cells,and protect HepG2 cells through the Keap1/Nrf2/HO-1 pathway.
