基于NLRP3炎症小体信号通路探讨补骨脂酚对膝骨关节炎软骨退变的影响
The Effect of Bakuchiol on Cartilage Degeneration in Knee Osteoarthritis Based on NLRP3 Inflammasome Signaling Pathway

Abstract
Objective:To investigate the effect of bakuchiol on cartilage degeneration in knee osteoarthritis(KOA)based on NOD-like receptor thermal protein domain associated protein 3(NLRP3)inflammasome signaling pathway.Methods:The KOA rat model was established by anterior cruciate ligament transection(ACLT).Thirty rats were randomly divided into a blank group,a model group and a bakuchiol group,with 10 rats in each group.The bakuchiol group was given bakuchiol 30 mg/kg·d by gavage,while the blank group and the model group were given an equal volume of normal saline by gavage for 4 consecutive weeks.Lequesne MG score was used to evaluate the behavior of the rats in various groups.ELISA method was used to detect the levels of serum interleu-kin-1β(IL-1β)and interleukin-18(IL-18)of the rats in various groups.Mankin score was used to evaluate the morphological changes of cartilage tissue after HE/safranin fast green staining.RT-qPCR was used to detect the mRNA expressions of NLRP3,apoptosis-associated speck-like protein containing a cartilage(ASC)and cysteine-aspartic proteases-1(Caspase-1)in cartilage tis-sue.Western Blot was used to detect the protein expressions of NLRP3,ASC,Caspase-1,aggrecan(ACAN)and type II collagen(Collagen II)in cartilage tissue.Results:Compared with the model group,bakuchiol could significantly reduce the Lequesne MG and Mankin scores of KOA rats(P<0.01);significantly reduce the levels of IL-1β and IL-18 in serum of KOA rats(P<0.01);signifi-cantly reduce the mRNA and protein expressions of NLRP3,ASC and Caspase-1 in cartilage tissue of KOA rats(P<0.01);and sig-nificantly increase the protein expressions of ACAN and CollagenII(P<0.01).Conclusion:Bakuchiol can improve KOA by regulat-ing the NLRP3 inflammasome signaling pathway,reducing the release of inflammatory factors and cartilage matrix degradation,and inhibiting cartilage degeneration.关键词
