摘要

Abstract

Objective:To observe the pain behavioral changes in rats with postherpetic neuralgia(PHN)treated with fire needle therapy,as well as changes in the expression of nerve injury-induced proteins 1,2(Ninjurin1,NINJ1;Ninjurin2,NINJ2)and pain me-diators in the dorsal root ganglia(DRG),to explore the analgesic mechanism of fire needle therapy.Methods:45 rats were randomly divided into a blank group,a model group,a solvent group(sham model),a fire needle group,and a gabapentin group,with 9 rats in each group.PHN rat models were prepared by intraperitoneal injection of resin toxin(RTX);After successful modeling,the fire needle group and gabapentin group were given corresponding intervention measures.After treatment,Western blotting was used to detect the expression levels of substance P(SP),calcitonin gene-related peptide(CGRP),glutamate(Glu),and NINJ1 and NINJ2 in the DRG of the 4th to 6th lumbar vertebrae of each group of rats,in order to explore the analgesic effect and mechanism of fire needle on PHN rats.Results:RTX intraperitoneal injection successfully established PHN models.After fire needle intervention,the mechanical pain threshold of PHN rats increased and the thermal pain threshold decreased,showing a continuous trend of change,in-dicating that fire needle has a significant analgesic effect;Under the intervention of fire needle,SP,CGRP,Glu,NINJ1,and NINJ2 in the dorsal root ganglia of rats were significantly reduced compared to the RTX group,indicating that the peripheral sensitization process of neuropathic pain was intervened.Conclusion:Fire needle therapy for PHN may regulate pain mediators and NINJ1 and NINJ2,intervene in the peripheral sensitization process to exert analgesic effects.
