温阳健脾法治疗阳虚体质人群的肠道菌群研究
Study on the Gut Microbiota of Individuals with Yang Deficiency Constitution Treated with the Method of Warming Yang and Strengthening Spleen

Abstract
Objective:To investigate the clinical effect of the method of warming Yang and strengthening the spleen in the treatment of people with Yang-deficient constitution clarify the effectiveness of the Warm Yang and Spleen Strengthening Method in treating people with Yang deficiency constitution,and explore the relevant mechanisms of the Warm Yang and Spleen Strengthening Method in improving Yang deficiency constitution from the perspective of gut microbiota.Methods:Forty volunteers with Yang deficiency constitution who received outpatient treatment at Guangdong Integrated Traditional Chinese and Western Medicine Hospital from January 2021 to June 2023 and met the inclusion criteria were selected and randomly divided into an experimental group and a con-trol group,with 20 cases in each group,using a random number table method.The control group received conventional treatment,while the experimental group received treatment with warming Yang and strengthening spleen(acupuncture+oral administration of Jin Kui Shen Qi Wan).Compare two groups of constitution transformation scores,and finally use 16S rDNA sequencing to deter-mine the alpha diversity,beta diversity,and relative abundance of gut microbiota at the phylum and genus levels in the patient's fe-ces.Results:After treatment,the transformation scores of Yang deficiency constitution in both groups of patients decreased signifi-cantly compared to before treatment(P<0.05),and the transformation scores in the experimental group were significantly lower than those in the control group after treatment(P<0.05).After treatment,the Shannon and Simpson values of both groups of patients were not statistically significant compared to before treatment(P>0.05),and the Chao1 index of the control group was not statistically sig-nificant compared to before treatment(P>0.05).However,the Chao1 index of the experimental group increased significantly com-pared to before treatment(P<0.05),and the Chao1 of the experimental group was significantly higher than that of the control group after treatment(P<0.05).The control group explained 6.80%and 6.77%of the community structure variation in PC1 and PC2 before and after treatment,respectively,and the distribution was relatively scattered along the PC1 and PC2 axes;The experimental group explained 8.19%and 7.63%of the community structure variation in PC1 and PC2 before and after treatment,respectively,and showed a clear separation trend in their distribution along the PC1 and PC2 axes.After treatment,the relative abundance of Fir-micutes,Bacteroidetes,Proteobacteria,and Desulfovibrio in the experimental group increased compared to before treatment(P<0.05),while the relative abundance of Verrucobacteria and Clostridium decreased compared to before treatment(P<0.05).There was no statistical difference in the relative abundance of Actinobacteria,Deinococcus,Cyanobacteria,and Campylobacter compared to before treatment(P>0.05).After treatment,the relative abundance of Bacteroides,Bifidobacterium,Fecal,and Koala genera in the experimental group increased compared to before treatment(P<0.05),while the relative abundance of Megamonas,Molluscum,Sal-monella,and Escherichia Shigella genera decreased compared to before treatment(P<0.05).There was no significant difference in the relative abundance of Prevotella and Klebsiella after treatment in the experimental group compared to before treatment(P>0.05).There was no statistically significant difference(P>0.05)in the levels of various bacterial phyla and genera before and after treat-ment in the control group.Conclusion:The treatment of warming Yang and strengthening spleen can improve the constitution of Yang deficiency,increase the richness of intestinal flora,and promote the relative aggregation of flora.This may be achieved by re-storing the structure of intestinal flora in Yang deficiency constitution to improve the constitution of Yang deficiency.关键词
