气流温湿度对鼻腔内流场的影响及多重因素对药物沉积规律的影响
The influence of airflow temperature and humidity on the flow field in the nasal cavity and the impact of multiple simulation on drug deposition patterns

Abstract
Objective To explore the influence of airflow temperature and humidity on the flow field in the nasal cavity,and analyze the effects of temperature,drug density,and drug particle size on the total deposition of drugs in the nasal cavity and the deposition in the olfactory region.To provide a basis for optimizing the nasal-brain targeted drug delivery device and improve the deposition rate of drugs in the olfactory region.Methods Firstly,the nasal cavity geometric model was constructed based on the CT scan images of healthy adults'nasal cavity.Then considering transient respiratory conditions,the flow field characteristics,including velocity field and pressure field distributions,were analyzed by using computational fluid dynamics methods under different combinations of temperature and humidity.What's more,the validation was carried out by comparing with the measured values of the nasal resistance.Then the variation law of drug deposition rate was investigated by using the response surface method considering the effects of the temperature,the particle size of the drug,and the density of the drug.Results In the anterior part of the nasal cavity,the airflow velocity increased with the increase of temperature,and the airflow pressure decreased with the increase of temperature.In the middle and posterior part of the nasal cavity,the airflow velocity and pressure inside the nasal cavity were not sensitive to the temperature of the inhaled airflow.The deposition rate of drug particles was significantly correlated with temperature,drug particle size,drug density,and the interaction of drug particle size and density.With the increase of temperature,the total deposition rate of the drug showed a trend of decreasing and then increasing.With the increase of drug particle size,the total deposition rate of drug particles showed a trend of decreasing and then increasing.When the temperature approached 25℃,the drug in the olfactory zone deposition rate was higher.The smaller the particle size of the drug was,the higher the deposition rate of the drug in the olfactory region was.When the drug particle size was small and the density of the drug was high,the deposition rate was low.When the drug particle size was large and the density of the drug was high,the deposition rate was high.Conclusions In order to achieve higher drug deposition rate in the olfactory zone,the ambient temperature can be maintained at 25℃,and the drug can be prepared into small size and low-density particles.关键词
