The influence of different bacterial suspensions on bacterial filtration efficiency experiments of masks

Abstract
Objective To explore the influence of Staphylococcus aureus strains from different sources(domestic strains and ATCC strains)and different diluents on the results of bacterial filtration efficiency(BFE)tests for masks,and to verify the feasibility of using domestic strains instead of ATCC strains as required by medical mask standards and using different diluents for BFE tests.Methods Three strains of Staphylococcus aureus from different sources[ATCC 6538,CICC 10306,CMCC(B)26003]were selected,and two diluents(physiological saline and 1.5%sterile peptone water solution)were used.Through confirmation tests on atomization volume,bacterial suspension concentration,positive quality control tests,and calculation of the mean particle size(MPS)of bacterial aerosols,the influence of different strain sources and diluents on BFE tests was analyzed and evaluated.Results Whether using physiological saline or a 1.5%sterile peptone water solution as the diluent for preparing bacterial suspensions,the colony-forming units in positive quality control tests ranged from 1700 to 3000 CFU,and the MPS values of bacterial aerosols ranged from 2.72 to 3.19 μm,meeting the standard test method requirements.There was no significant difference in BFE test results for the three strains diluted with different diluents(P=0.108).Conclusions This study verifies the feasibility of using strains other than ATCC 6538 Staphylococcus aureus strains required by medical mask standards for BFE tests and can choose physiological saline as the diluent for preparing bacterial suspensions,greatly simplifying the test operation process,reducing test costs,and providing more convenient test schemes for production enterprises and inspection agencies,expanding the operability of BFE tests in practical applications.Through exploring relevant tests of domestic biotechnology innovation,we hope to provide scientific basis for the update of industry standards.关键词
