Effect of lipoic acid-functionalized gold nanoparticles in promoting muscle regeneration and functional recovery

Abstract
Objective To investigate the effects of lipoic acid-functionalized gold nanoparticles(LA-Au NPs)on the antioxidant capacity of myoblasts,and to evaluate their potential in promoting muscle regeneration and functional recovery,thereby providing a novel therapeutic strategy for sarcopenia.Methods Lipoic acid-functionalized gold nanoparticles(LA-Au NPs)of different sizes were synthesized using the chloroauric acid reduction method and characterized by transmission electron microscopy.The cytotoxicity of LA-Au NPs on mouse myoblast C2C12 cells was assessed using the Alamar Blue assay.An oxidative stress microenvironment was simulated using hydrogen peroxide(H2O2)to investigate the effects of LA-Au NPs on cell viability,apoptosis,and intracellular reactive oxygen species(ROS)production in myoblasts.Furthermore,the therapeutic effects of LA-Au NPs on body weight,muscle mass,strength,and exercise capacity were evaluated in a sarcopenia mouse model.Results Four types of LA-Au NPs with different sizes were successfully synthesized,with diameters of(3.52±0.36)nm,(26.99±2.01)nm,(57.93±5.61)nm,and(96.29±7.72)nm,all exhibiting uniform spherical morphology.LA-Au NPs showed no significant cytotoxicity to C2C12 cells within the concentration range of 0-50 μg/mL,indicating good biocompatibility.Under H2O2-induced oxidative stress,LA-Au NPs significantly reduced intracellular ROS levels and inhibited cell apoptosis,with smaller LA-Au NPs-4 demonstrating stronger protective effects.In the sarcopenia mouse model,treatment with LA-Au NPs-4 significantly improved body weight,muscle mass,grip strength,motor coordination,and endurance,indicating their potential to promote muscle regeneration and functional recovery.Conclusions LA-Au NPs enhance the antioxidant capacity of myoblasts and significantly promote muscle regeneration and functional recovery.These findings highlight the potential therapeutic application of LA-Au NPs in the treatment of sarcopenia.The study provides new insights into the application of nanotechnology in sarcopenia therapy and lays the foundation for the further development of LA-Au NPs in the biomedical field.关键词
