Abstract

Objective To observe the effect of Shenqi Guiteng Decoction on the inflammatory pathway of nuclear transcription factor-κB(NF-κB)in IgA nephropathy rats,and to explore its mechanism of action in the treatment of IgA nephropathy.Methods 42 rats were randomly divided into the normal group(n=10),the model group(n=12),the benazepril group(n=10),and the Shenqi Guiteng Decoction group(n=10).Except for the blank group,in the other groups IgA nephropathy rat models were established by gavage of bovine serum albumin(BSA)(400 mg·kg-1),subcutaneous injection of castor oil(0.5 mL)and carbon tetrachloride(CCl4)(0.1 mL),and tail vein injection of lipopolysaccharide(LPS)(0.05 mg).After 13 weeks of modeling,the benazepril group was treated with benazepril hydrochloride tablets by gavage for 4 weeks,while the Shenqi Guiteng Decoction group was treated with Shenqi Guiteng Decoction by gavage for 4 weeks.After the end of administration,the 24-hour urine protein,total protein(TP),albumin(ALB),and urea(UREA)levels in each group were detected.Hematoxylin and eosin(HE)staining,periodic acid Schiff(PAS)staining,and Masson staining were used to observe renal pathological changes.Immunofluorescence was used to observe the deposition of IgA in the glomerular mesangial membrane,transmission electron microscopy was used to observe the ultrastructure of renal tissue,and protein immunoblotting was used to detect the expression of NF-κB p-65(p65),phosphorylated p65(p-p65),and interleukin-1 β(IL-β)proteins in renal tissue.Results Compared with the normal group,the body mass and ALB of the model group significantly decreased before and after administration(P<0.01),24-hour urine protein quantification,UREA,glomerular tethered membrane cell proliferation,deposition of tethered membrane IgA and electron-dense material,and p-p65 and IL-1β protein expression in renal tissues all increased(P<0.01);Compared with the model group,the levels of ALB and UREA in the benazepril group and the Shenqi Guiteng Decoction group were significantly increased and decreased respectively(P<0.05),and the pathological indicators of renal tissue improved.The 24-hour urine protein and the protein expressions of p-p65 and IL1β in renal tissue were significantly decreased(P<0.01).Conclusion Shenqi Guiteng Decoction can effectively treat IgA nephropathy,and its mechanism of action may be related to inhibiting the activation of NF-κB inflammatory and reducing glomerular inflammation damage.
