摘要

Abstract

Objective To investigate the effects of Taipei Fritillary Bulb(TFB)on the proliferation and migration of hepatocellular carcinoma HepG2 and Bel-7402 cells and its mechanism.Methods 50 SD rats were randomly divided into the blank control group,TFB low(0.5 g·mL-1)group,TFB medium(1 g·mL-1)group,TFB high(2 g·mL-1)group and cisplatin positive control group(1 mg·mL-1).Drug-containing serum was prepared after 7 days of intragastric administration for the culture of HepG2 and Bel-7402 cells.Cell proliferation ability was detected by CCK-8 assay.Cell migration ability was detected by scratch test.Western Blot assay was used to detect expression levels of cysteine aspartic protease Cleaved Caspase-3 and Cleaved Caspase-9,Blymphocyte tumor(Bcl)-2 related proteins Bax,Bcl-2,PI3K,AKT,human phosphorylated p-PI3K,and p-AKT protein;The relative expression levels of PI3K and AKT mRNA were detected by RT-qPCR.Results TFB might inhibit proliferation and migration of HepG2 and Bel-7402 cells(P<0.05,P<0.01),increase protein expression of Bax,Cleaved Caspase-3,Cleaved Caspase-9,and decrease the expression of Bcl-2 and the relative expression of PI3K and AKT mRNA(P<0.05,P<0.01).Conclusion TFB can inhibit the proliferation and migration of hepatocellular carcinoma cells by regulating PI3K/AKT signaling pathway.
