解毒化瘀泄肺方对脂多糖致大鼠急性肺损伤的治疗作用及机制研究
Study on the therapeutic effect and mechanism of Jiedu Huayu Xiefei Prescription on acute lung injury induced by lipopolysaccharide in rats

Abstract
Objective To explore the therapeutic effect and mechanism of Jiedu Huayu Xiefei Prescription on acute lung injury(ALI)induced by lipopolysaccharide in rats.Methods 60 SD rats were randomly divided into the normal group,the model group,the JDHYXF low-dose group,the JDHYXF high-dose group,and the Dexamethasone group,with 12 rats in each group.Except for the normal group,all other groups used the method of tracheal instillation of lipopolysaccharide(LPS)to establish an ALI rat model.Lung tissue was taken,and the wet-dry weight(W/D)ratio of lung tissues was measured.The inflammatory factors and oxidative stress indexes were detected by enzyme-linked immunosorbent assay(ELISA).Hematoxylin eosin(H&E)staining was used to analyze the pathological damage.The expression of related proteins was detected by Western blot.Results Compared with the normal group,W/D ratio,and the levels of interleukin(IL)-6,IL-1β,tumor necrosis factor-α(TNF-α),malonaldehyde(MDA),myeloperoxidase(MPO),vascular endothelial growth factor(VEGF),hypoxia-inducing factor-1α(HIF-1α),phosphoinositol 3 kinase(PI3K),protein kinase(AKT),and mammalian Rapamycin target protein(mTOR)in the model group were significantly increased(P<0.05),while the levels of total superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were decreased(P<0.05).Compared with the model group,the W/D ratio,and the levels of IL-6,IL-1β,TNF-α,MDA,MPO,VEGF,HIF-1α,PI3K,AKT,and mTOR proteins were significantly decreased in the Dexamethasone group,the JDHYXF low-dose group,and the JDHYXF high-dose group(P<0.05),while SOD and GSH Px levels were increased(P<0.05).Moreover,LPS-induced lung tissue injury and inflammatory cell infiltration were significantly decreased.Conclusion Jiedu Huayu Xiefei Prescription significantly improve the inflammatory and oxidative stress levels of ALI induced by lipopolysaccharide in rats.The regulation of PI3K/AKT/mTOR/HIF-1α/VEGF signaling pathway may be the key mechanism of the protective effect of Jidu Huayu Xiefei Prescription on ALI treatment.关键词
