Abstract

Objective:To investigate how Wenxin granules regulate energy metabolism in post-myocardial infarction(MI)heart failure rats via the peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α)/peroxisome proliferator-activated receptor alpha(PPARα)signaling pathways.Methods:The rat model of post-myocardial infarction heart failure was es-tablished by ligating the left anterior descending coronary artery.Subsequently,the rats were randomly categorized into the sham operation group,model group,low-dose Wenxin Granules group,high-dose Wenxin Granules group,and trimetazidine group.Af-ter 4 weeks,cardiac function was assessed by echocardiography,and cardiac troponin T(cTnT)levels were measured using ELI-SA.ATP,AMP,and ADP levels were determined,and ADP/ATP and AMP/ATP ratios calculated.Mitochondrial respiratory chain complexⅠ、Ⅱ、Ⅳ、Ⅴ activities were evaluated.Western blotting was used to assess PGC-1α,PPARα,carnitine palmitoyl-transferase 1α(CPT1α)and fatty acid transport protein(CD36).Results:Compared with the sham operation group,the cardiac function parameters of the model group significantly decreased.The structure-related parameters showed pathological characteris-tics of thinner ventricular walls and enlarged heart cavities.The level of cTnT was significantly increased,along with reduced re-spiratory chain complex activities and ATP levels,and increased AMP and ADP levels.Protein expressions of PGC-1α,PPARα,CD36,and CPT1α were significantly downregulated.Compared with the model group,the above detection indexes of the Wenxin granules group showed varying degrees of improvement after intervention.Conclusion:Wenxin granules,by modulating the PGC-1α/PPARα signaling pathway,enhance energy metabolism and confer cardioprotection on rats with post-myocardial infarction heart failure.
