基于TLR4/MyD88/NF-κB信号通路研究平喘颗粒对咳嗽变异性哮喘小鼠气道炎症的作用机制
Mechanism of Pingchuan Granules on airway inflammation in mice with cough variant asthma based on TLR4/MyD88/NF-κB signaling pathway

Abstract
Objective:To investigate the mechanism of Pingchuan Granules in reducing cough variant asthma(CVA)airway inflammation by exploring the expression of TLR4,MyD88,and NF-κB in lung tissue of mice with CVA.Methods:A total of 60 mice were randomly divided into the blank group,model group,Pingchuan Granules group,and positive control group(montelu-kast sodium group)using the number table method.Intraperitoneal injection of sensitization solution(100 μg OVA+20 μg alumi-num hydroxide)combined with nebulization stimulation(1.5%OVA)was used to build a CVA model.At the same time,differ-ent groups were given Pingchuan Granules,montelukast sodium,and physiological saline by gavage.H&E staining was used to observe the morphology of mice lung tissues;enzyme linked immunosorbent assay was used to detect the levels of IL-4 and IL-13 in the serum of mice,immunohistochemical staining was used to detect TLR4 and NF-κB protein expression,and Western blot was used to detect TLR4,MyD88,and NF-κB protein expression levels.Results:Compared to the blank group,the model group mice showed more infiltration of inflammatory cells in the lung tissues,incomplete bronchial wall structure,thickening of ep-ithelial areas,and significant expansion of alveoli;TLR4,MyD88,and NF-κB protein were significantly increased in mice lung tissues(P<0.05,P<0.001);the levels of IL-4 and IL-13 were significantly increased in the serum of mice(P<0.001).Com-pared to the model group,the Pingchuan Granules group and the positive control group showed reduced infiltration of inflammato-ry cells in lung tissues,reduced thickness of epithelial sites,and intact morphology of bronchial wall structure;the relative expres-sion levels of TLR4,MyD88,and NF-κB proteins in the lung tissues of mice were significantly increased(P<0.05,P<0.001);the levels of inflammatory factors IL-4 and IL-13 in the serum of mice were significantly downregulated(P<0.05,P<0.001).Conclusion:Pingchuan Granules can down-regulate the expression levels of TLR4,MyD88,and NF-κB in mice lung tissues,fur-ther inhibit the activation of TLR4/MyD88/NF-κB signaling pathway,and effectively improve airway inflammation in CVA mice.关键词
