Abstract

Objective:To investigate the expression of tyrosine kinase with immunoglobulin-like and EGF-like domains 1(TIE1)in human tumor tissues and determine its potential clinical significance.Methods:Various online websites and databases,such as TIMER2,GEPIA2,cBioPortal,UALCAN,and STRING,were used to explore the relationship between TIE1 expres-sion and prognosis,genetic alterations,tumor immunity,and methylation status across different types of cancer.This research in-volved the examination of TIE1 expression through a dataset consisting of patients diagnosed with cervical cancer.The molecular mechanisms underlying TIE1 were explored through Kyoto Encyclopedia of Genes and Genomes(KEGG)as well as Gene Ontol-ogy(GO)enrichment analyses.Additionally,in vitro studies were conducted using wound healing,and transwell assays to deter-mine the biological function of TIE1 in cervical cancer cells.Results:Pan-cancer analysis revealed upregulation of TIE1 in certain tumors,and its expression was significantly associated with the prognosis of cancer patients.We examined the gene alteration sta-tus of TIE1 in various tumor samples and found that patients with skin melanoma(skin cutaneous melanoma,SKCM)had the highest frequency of gene alterations.Furthermore,TIE1 expression is closely linked to immune cell infiltration in some tumors.Significant differences were observed in the promoter methylation levels of TIE1 in certain tumors.The analysis of KEGG enrich-ment indicated that genes associated with TIE1 were significantly represented in the signaling pathways of Ras,PI3K-Akt,RAP1,and MAPK.Similarly,The GO enrichment analysis indicated that pathways associated with externally wrapped structural tissues were enriched with genes related to TIE1,collagen-containing extracellular matrix,and glycosaminoglycan binding.Experi-ments conducted in vitro additionally validated that TIE1 enhances the movement and infiltration of cervical cancer cells.Conclu-sion:The expression of TIE1 showed a correlation with clinical outcomes,genetic mutations,and the infiltration of immune cells in tumors.This study provides the critical fundamental data to understand the role of TIE1 in tumorigenesis and development.
