Physiological and Biochemical Properties During Development of Pinus massoniana Twigs and the Relationship with Rooting

Abstract
[Objective]The relationships between intrinsic physiological and biochemical properties and rooting during the development stage of Pinus massoniana twigs were explored,and the appropriate rooting time from perspective of internal quality of cuttings,which provided theoretical basis for applying P.massoniana cuttings in production.[Method]Using P.massoniana cuttings as materials,the rooting effects of cuttings collected in different months were compared,and the contents of four endogenous hormones,including indole-3-acetic acid(IAA),abscisic acid(ABA),zeatin(ZT)and gibberellin(GA),as well as the differences in soluble sugar and soluble protein,were detected and analyzed.The activity differences of five enzymes,including peroxidase(POD),catalase(CAT),polyphenol oxidase(PPO),superoxide dismutase(SOD),and indolyl acetic acid oxidase(IAAO),during the development of cuttings were analyzed,revealing the relationship between the physiological and biochemical characteristics of cuttings in different months and rooting.[Result]The nutrients,endogenous hormones and related enzymes activities in P.massoniana cuttings varied from month to month.The rooting rate of P.massoniana cuttings varied from January to December,with higher rates in October to December and lower rates in May to July.The rooting rate was positively correlated with the soluble sugar content,carbon to nitrogen ratio(C/N),was very significantly and positively correlated with IAA content,POD activity and IAA/ABA,and was negatively correlated with soluble protein content.[Conclusion]From October to December,the C/N ratio of the panicles was relatively high.The internal nutrients of the panicles are abundant,the lignification degree is high,the stress resistance is strong,and the survival rate is high.IAAO promotes the formation of adventitious root by regulating IAA content.It is advisable to choose cutting from October to December to achieve a high survival rate.关键词
