Physiological Response of Dendrobium officinale from Different Regions Under Low Temperature Stress and Evaluation on Its Cold Resistance

Abstract

[Objective]The physiological index change of Dendrobium officinale from different regions under low temperature stress was explored,and the key indicators influencing cold resistance were identified,which provided references for promoting its cultivation in suitable regions.[Method]D.officinale samples from different regions were subjected to low temperature treatments at 4℃,0℃,-4℃,and-8℃.Physiological traits including soluble protein(SP),malondialdehyde(MDA),chlorophyll(Chl)content,and activities of three antioxidant enzymes[superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)]were measured.The LT50 was evaluated by fitting relative electrical conductivity(REC)under different temperatures.Variance analysis,correlation analysis,and principal component analysis(PCA)were conducted to study cold resistance mechanisms.[Result]LT50 of D.officinale from different regions followed GZY-K(-0.484 6℃)>GZY-080(-0.516 3℃)>GZY-003(-2.241 0℃)>GZY-Y(-3.299 1℃),with GZY-Y exhibiting the strongest cold resistance.At-4℃,severe frost damage occurred,and the cold resistance system collapsed.Extremely significant differences in physiological index of D.officinale from different regions were observed among cultivars and temperature treatments,indicating rich genetic diversity in cold resistance.Under low temperature stress,physiological index existed complex correlation.CAT activity showed a highly significant negative correlation with SOD activity and EC,but a significant positive correlation with POD activity.MDA content was highly and negatively correlated with POD activity and significantly and negatively correlated with Chl content.Chl content was highly and positively correlated with SP.SPs was highly and negatively correlated with POD and SOD activities,and significantly and negatively correlated with EC.At 0℃,SOD activity,Chl content,SP,and POD activity synergistically or antagonistically determined cold resistance.[Conclusion]Cold resistance of D.officinale from different regions follows GZY-Y>GZY-003>GZY-080>GZY-K,with GZY-Y being the most cold-tolerant,suitable for breeding cold-resistant cultivars.Cold resistance mechanisms vary significantly among regional D.officinale under low temperature stress.SOD activity,Chl content,SP,and POD activity are critical traits for cold resistance formation of D.officinale.
