莙荙菜叶绿体基因组结构与特征分析
Analysis of Structure and Characteristics of Chloroplast Genome of Chard(Beta vulgaris var.cicla L.)

Abstract
[Objective]The characteristics of the chloroplast genome of chard and its phylogenetic relationships were studied to provide theoretical foundation for species identification,genetic evolution research and variety improvement of chard.[Method]Chard varieties from Guizhou Province were selected as the research materials.Illumina high-through-put sequencing technology was utilized for sequencing.Subsequently,the GetOrganelle software was employed for sequence assembly,and the sequences were annotated through Geseq and web servers.CodonW1.4.2 software was used to analyze codon bias,and the MISA online software was adopted to identify the simple sequence repeat(SSR)loci.Mafft and fasttree were used to construct phylogenetic trees.[Result]The chloroplast genome of chard was successfully assembled.The complete chloroplast genome of chard was 149 703 bp long,presenting a typical double-stranded circular quadripartite structure.It contained 131 genes and 64 types of codons.Except UAA,UAG and UGA,other codons all belonged to stop codons,and the other codons constituted 20 amino acids of protein.Among them,leucine exhibited the highest coding rate,and most of the frequently used synonymous codons terminated with A or U.Additionally,a total of 59 SSR loci were identified,with the number of mononucleotide repeats(A/T)being the largest.The phylogenetic analysis results indicated that chard had a relatively close genetic relationship with sugar beet and spinach.[Conclusion]The chloroplast genome of chard is 149 703 bp in length,and has the most closely genetic relationship with sugar beet.关键词
