摘要

Abstract

Objective:To investigate the mechanism of Wutou Decoction in delaying cartilage degeneration of knee osteoarthritis(KOA)by regulating the P38 MAPK signaling pathway.Methods:Sixty 8-week-old SD male rats were acclimatised and fed for 1 week and then divided into normal group(n=15)and modeling group(n=45)using a random number table.The modeling group was used to establish a rat KOA model using the modified Hulth method.After the model was successfully established,the modeling group was further divided into model group,treatment group,and control group(n=15)using random numbers.The treatment group was given 4.2 g/mL of Wutou Decoction at a body mass of 10 mL/(kg·d)by gavage,while the control group was given 1 mg/mL di-clofenac sodium solution at the same dose by gavage.The normal and model groups were given an equal dose of saline by gavage.Af-ter an 8-week continuous intervention,the four groups of rats were executed under anesthesia,and the cartilage tissues of the bilateral knee joints were separated and collected.The HE staining method was utilised to observe the morphology of cartilage tissues in the four groups.The related proteins of P38 mitogen-activated protein kinase(P38 MAPK),c-Jun N-terminal kinase(JNK)rat sarcoma(Ras),phosphorylated extracellular-regulated protein kinase 1/2(p-ERK1/2),and inducible nitric oxide synthase(iNOS)were detected by Western blot in the cartilage tissues of the four groups.Results:The cartilage tissue structure in the normal group showed continu-ity and integrity,with intact tangential layers and no signs of defects.Compared with the normal group,the cartilage tissue in the mod-el group showed disrupted and discontinuous superficial structure in the tangential layer,with fiber direction deviation and thinning.Some chondrocytes in the transitional and radial layers were disordered and irregularly arranged.The protein expression levels of P38 MAPK,JNK,Ras,p-ERK1/2 and iNOS in cartilage tissues all increased(P<0.05).Compared with the model group,the morpho-logical structure in the superficial layer of the treatment and control groups was relatively complete,with no defects in the tangential layer,and the structures of other layers in the cartilage tissue were clear and distinguishable.The protein expression levels of P38 MAPK,JNK,Ras,p-ERK1/2,and iNOS in cartilage tissues all decreased(P<0.05).Conclusion:Wutou Decoction can enhance the morphology of articular cartilage in rats with KOA,and alleviate KOA symptoms.The mechanism of action of Wutou Decoction in-volves the regulation of the expression levels of proteins related to the P38 MAPK signaling pathway.
