摘要

Abstract

Based on the mechanism by which RecE/RecT(RecET)mediates short homology arms(30-60 bp)DNA recombination,this paper explores the application prospects of RecET recombination as a new tool widely used in plasmid construction and genome editing of metabolic pathway.The develop-ment of the recombination tool from construction to perfection,its application in plasmid construction and genome engineering,crystal and cryo-electron microscopy structure of the RecE/RecT proteins,and mechanism hypothesis of exonuclease activity and annealing of ssDNA were reviewed.Further-more,the existing problems and future development of in vitro recombination,in vitro interaction,and research value in eukaryotes were prospected.
