Abstract

[Objective]This study aimed to screen microbial strains capable of efficiently degrading tobacco leaf proteins,study their enzymatic properties of proteases,and explore proteases with indus-trial application value.[Method]The strains with high protein degradation efficiency were isolated from the surface of tobacco leaves by protease selective medium and enzyme activity determination.The morphological,molecular biology,physiological and biochemical identification of the screened strains were carried out,and the characteristics of the protease produced(temperature,pH value,metal ion)were further studied to explore its effect on protein degradation of tobacco leaves.[Result]A bacterial strain YANDB-21 with high efficiency protein degradation was screened and identified as Stenotrophomonas maltophilia.The protease produced by the bacteria has enzyme activity under acidic,neutral and alkaline conditions.The optimum reaction temperature of the protease was 50℃,and the optimum reaction pH was 10.0.The enzyme has good temperature tolerance below 40℃,and good stability in the pH 3.0~12.0.The metal ions K+,Cu2+,Ca2+and Mn2+can improve the enzyme activity,while Mg2+,Zn2+,Fe2+and Fe3+inhibit the enzyme activity.The crude enzyme solution was used to treat Shaanxi Shangluo tobacco leaves,and the protein degradation rate was 22.42%.[Conclu-sion]The protease enzyme produced by strain YANDB-21 has high activity and good stability,and can efficiently degrade tobacco leaf protein,which is suitable for promotion and application in tobacco pro-cessing and production.
