水稻OsIAA10蛋白的表达及纯化
Expression and Purification of OsIAA10 in Rice
摘要
Abstract
OsIAA10 plays an important role in the growth,development,and stress responses of rice.This study aims to explore the biochemical nature of OsIAA10.With the cDNA of wild-type'Zhonghua 11'(ZH11)plants as the template,and the coding sequence of OsIAA10 was amplified with the forward and reverse primers of pGEX-6p-1-IAA10.Then,the recombinant plasmid pGEX-6p-1-IAA10 containing OsIAA10 was obtained through vector construction and transferred into BL21 cells.After IPTG-induced expression,purification,and SDS-PAGE detection,a single protein band consistent with the expectation was obtained,and it was verified to be OsIAA10.The induction conditions for the expression of this protein were then optimized.The results showed that the treatment with 0.2 mmol/L IPTG at 37℃for 12 h were the optimal conditions for inducing the expression of OsIAA10.The research results underpin the subsequent research on the function of OsIAA10.关键词
