刺五加皂甙靶向RIPK2抑制肺腺癌细胞增殖的机制研究

Abstract
Objective To analyze the mechanism of acanthopanax senticosus saponins(ASS)inhibiting the proliferation of lung adenocarcinoma(LUAD)cells by targeting receptor-interacting serine/threonine kinase 2(RIPK2).Methods PC-9 and A549 cell lines were cultured and divided into the control group,the ASS group,the RIPK2 overexpression group,and the ASS+RIPK2 overex-pression group.The survival rates of cells under different concentrations of ASS were detected by CCK-8.The number of clone cells was detected by clone formation assay.The number of PC-9 and A549 cells,and their apoptosis rates in each cell cycle were detected by flow cytometry.The body mass of xenograft tumors at different time points were observed.The expression of Ki67 was detected by immu-nohistochemistry.The expression levels of RIPK2 in each group were detected by Western Blot.Results The expression level of RIPK2 was related to depth of tumor invasion,clinical staging,lymph node metastasis,recurrence,prognosis,and survival rate.The higher the concentration of ASS,the lower the survival rates of PC-9 and A549 cells,and the number of clone cells(P<0.05).The volume and weight of xenograft tumors,and expression of Ki67 in ASS group were significantly lower than those in the control group(P<0.05).With the increase of ASS'action time and concentration,the expression of RIPK2 in PC-9 and A549 cell lines was significantly decreased(P<0.05).Compared with the control group,number of PC-9 and A549 cells in G0/G1 phase,and apoptosis rate were sig-nificantly increased in the ASS group,while number of cells in S phase,expression level of RIPK2,activities of PC-9 and A549 cells,and the number of clone cells were significantly decreased(P<0.05).Compared with the control group,number of cells in S phase,expression level of RIPK2,activity of cancer cells and number of clone cells were significantly increased in RIPK2 group,while number of PC-9 and A549 cells in G0/G1 phase,and apoptosis rate were significantly decreased(P<0.05).Compared with the ASS group,number of cells in S phase,expression level of RIPK2,activity of cancer cells and number of clone cells were significantly increased in the ASS+RIPK2 group,while number of PC-9 and A549 cells in G0/G1 phase,and apoptosis rate were significantly decreased(P<0.05).Conclusion ASS may inhibit the malignant proliferation of LUAD cells by down-regulating the expression level of RIPK2.关键词
