摘要

Abstract

To optimize solid fermentation conditions of Metarhizium anisopliae M14-1,the biocontrol fungus against of Monochamus saltuarius,and improve fermentation efficiency and spore yield,single factor experiment was used to determine parameters of solid medium and optimal culture conditions for strain M14-1,and important influencing factors were screened by Plackett-Burman experiment.On the basis,response surface method was used to optimize the important factors.The results showed that optimal conditions for solid fermentation of M.anisopliae M14-1 were as follows:mass ratio of rice flour to rice husk was 1.52∶1.95,glucose 0.10%,ammonium sulphate 0.10%,magnesium sulfate 0.10%,water content 80%,loading amount 100 g,inoculation amount 15%,fermentation temperature 27.18℃,fermentation time for 13 d.Spore production of strain M14-1 was 1.29 billion spores/g after optimization,but before optimization,spore production was 0.588 billion spores/g,and spore production effect was significantly improved.Solid fermentation conditions of strain M14-1 was optimized,and the results can provide theoretical basis for practical application.
