目的 探讨富血小板血浆(PRP)对皮肤细胞增殖和分化的影响及其作用机制.方法 体外培养人真皮成纤维细胞(HDFs)和人表皮角质形成细胞(HEKs),并给予不同浓度的PRP处理;采用CCK-8法、免疫荧光染色、ELISA等技术,对细胞增殖、分化及相关生长因子表达进行检测,系统评估不同浓度PRP对HDFs和HEKs的影响.结果 PRP能显著提高HDFs和HEKs的增殖率,促进细胞分化,并上调血小板源性生长因子(PDGF)、表皮生长因子(EGF)、成纤维细胞生长因子(FGF)表达,差异具有统计学意义(P<0.05).结论 在促进皮肤细胞增殖和分化方面,PRP可提高生长因子的表达水平.

Objective To explore the effects of platelet-rich plasma(PRP)on the proliferation and differentiation of skin cells and its mechanisms of action.Methods Human dermal fibroblasts(HDFs)and human epidermal keratinocytes(HEKs)were cultured in vitro and subjected to varying concentrations of PRP at different concentrations.Cell counting kit-8(CCK-8),immunofluo-rescence staining and enzyme-linked immunosorbent assay(ELISA)techniques were utilized to evaluate cell proliferation,differentia-tion,and the expression of associated growth factors,and the effects of different concentrations of PRP on HDFs and HEKs were sys-tematically evaluated.Results PRP significantly enhanced the proliferation rates of HDFs and HEKs,facilitated cell differentiation,and upregulated the expression of growth factors such as platelet derived growth factor(PDGF),epidermal growth factor(EGF),and fi-broblast growth factor(FGF),and the differences were statistically significant(P<0.05).Conclusions PRP can increase the expres-sion level of growth factors in promoting the proliferation and differentiation of skin cells.
