目的:鉴定男性生殖功能相关的开放阅读框基因编码蛋白C3ORF20 的调节因子,并探究其生殖调控功能.方法:通过人体组织样本RNA-seq数据和人睾丸样本scRNA-seq数据分析C3ORF20 的时空表达;生物信息学分析C3ORF20 的互作蛋白并构建PPI网络;KEGG/GO分析互作蛋白参与的信号通路及分子事件;scRNA-seq分析及RT-PCR实验验证BIRC6与C3ORF20的时空共表达;Co-IP和GST-pull down检测蛋白相互作用及精细互作区段;泛素化实验和免疫共沉淀实验检测BIRC6 对C3ORF20 蛋白稳定性的影响.结果:C3ORF20 mRNA在早期和中期圆形精子细胞中表达;C3ORF20与凋亡抑制蛋白家族成员BIRC6 存在相互作用;BIRC6 通过其泛素结合酶结构域UBC与C3ORF20蛋白的 FAM194 结构域相互作用;BIRC6 蛋白泛素化 C3ORF20 并导致其不稳定.结论:BIRC6 是C3ORF20蛋白的一个功能调节因子,其通过泛素化途径调节C3ORF20蛋白的稳定性,进而影响其生殖调控功能.

Objective To identify the regulatory factors associated with the reproduction-related open reading frame gene C3ORF20 and explore its regulatory function in reproduction.Methods The spatiotemporal expression of C3ORF20 was an-alyzed using RNA-seq data from human tissue samples and scRNA-seq data from human testis samples.Bioinformatics was used to identify interacting proteins of C3ORF20 and a protein-protein interaction(PPI)network was constructed.KEGG/GO analysis was conducted to explore the signaling pathways and molecular functions involving these interacting proteins.The co-expression of BIRC6 and C3ORF20 was verified through scRNA-seq analysis and RT-PCR.Co-immunoprecipitati-on(Co-IP)and GST pull-down assays were employed to detect protein interactions and fine interaction domains.Ubiquiti-nation and immunoprecipitation were carried out to analyze the effect of BIRC6 on the stability of C3ORF20.Results C3ORF20 mRNA was specifically expressed in early and mid-round spermatids.Interaction between C3ORF20 and BIRC6 was observed,and BIRC6 interacted with the family with sequence similarity 194(FAM194)domain of C3ORF20 through its ubiquitin-conjugating enzymes(UBCs).BIRC6 ubiquitinated C3ORF20,which results in its instability.Conclusion BIRC6,as a regulator of C3ORF20,regulates its stability through the ubiquitination pathway,thereby affecting its regulato-ry function in reproduction.
