Studies of the CERK1 F322 Locus Affecting Innate Immunite Response in Arabidopsis

Abstract
Objective To study the function of conserved amino acids in the juxtamembrane domain of CERK1 in Arabidopsis thaliana in mediating chitin-induced immune responses.Methods Bioinformatics was employed to analyze the conservation of amino acids in the LysM receptor family of A.thaliana.Mutants of CERK1 and conserved amino acid residues in the in-tracellular juxtamembrane domain were constructed and expressed transiently in Nicotiana benthamiana to verify their im-mune functions.Using Arabidopsis transformation technology,the roles of these conserved amino acid residues were investi-gated in mediating chitin-induced immune responses through quantitative fluorescence assay and Western blot in conserved amino acid mutations in juxtamembrane domain of CERK1.Results Amino acid sequences in the juxtamembrane domain of the LysM receptor kinase family of A.thaliana were analyzed,which identified three conserved amino acid sites in CERK1:T319,F322,and S325.The overexpression of CERK1F322A resulted in the disability of inducing hypersensitive response in N.benthamiana.In CERK1F322A transgenic lines,the transcriptions of WRKY33 and WRKY53 in response to chitin were sup-pressed,along with the phosphorylation modification of MAPK3/6.Conclusion The F322 locus is a critical amino acid site for CERK1 in transducing immune signals,highlighting its significant role in understanding the immune regulation by the juxtamembrane domain of CERK1 and the transduction of immune signals.关键词
