目的:本研究旨在通过生物信息学和免疫组化染色(IHC)分析RAB3B基因在肝细胞癌(LIHC)中的表达模式及其与诊断、预后和肿瘤细胞糖酵解的相关性.方法:利用TCGA和GEO数据库中的数据,通过差异基因筛选、相关性分析、诊断效能评估、预后分析、功能富集分析和糖酵解相关性分析等步骤,探讨RAB3B在LIHC中的作用.通过IHC染色验证 RAB3B的表达差异.结果:RAB3B 在 LIHC 肿瘤样本中的表达显著高于正常样本(P<0.05),并展现出较高的诊断效能(TCGA LIHC;AUC:0.878,95%CI=0.841~0.916).RAB3B表达水平与患者的总体生存(OS)和无复发存活率(RFS)密切相关(P<0.05).IHC结果证实肿瘤样本中RAB3B表达显著高于癌旁样本(P<0.05).功能富集分析表明RAB3B与 329 个基因相关,涉及糖蛋白生物合成及内质网蛋白质加工等过程.此外,RAB3B表达与糖酵解关键基因GPI的mRNA表达水平呈正相关(P<0.05).结论:RAB3B的高表达与 LIHC患者的进展、预后和糖酵解密切相关,它可能成为 LIHC预测预后的生物标志物和治疗靶点.

Objective To analyze the expression pattern of RAB3B in hepatocellular carcinoma(LIHC)and its correlation with diagnosis,prognosis,and tumor cell glycolysis using bioinformatics and immunohistochemical(IHC)staining.Meth-ods The data from TCGA and GEO databases were utilized to explore the role of RAB3B in LIHC through differential gene screening,correlation analysis,diagnostic efficacy assessment,prognostic analysis,functional enrichment analysis,and glycolysis correlation analysis.The expression differences of RAB3B were verified by IHC staining.Results The expression of RAB3B was higher in tumor samples than that in normal samples(P<0.05)and it displayed high diagnostic efficacy(TCGA LIHC;AUC:0.878,95%CI=0.841~0.916).The expression of RAB3B was closely correlated with overall surviv-al and relapse-free survival of patients(P<0.05).IHC staining confirmed that RAB3B in tumor samples was higher than that in adjacent normal tissues(P<0.05).Functional enrichment analysis indicated that RAB3B was associated with 329 genes involved in glycoprotein biosynthesis and endoplasmic reticulum protein processing.Moreover,RAB3B was positively correlated with the mRNA expression of the glycolysis key gene GPI(P<0.05).Conclusion The high expression of RAB3B is closely related to the progression,prognosis,and glycolysis of LIHC patients,suggesting it may serve as a biomarker for predicting the prognosis and therapeutic target in LIHC.
