Abstract

Objective To explore the effect of edaravone dexcamphene on the changes of inflammatory factors in protecting cerebral ischemia-reperfusion injury by regulating the TLR4/NF-κB signaling pathway.Methods Hippocampal neuron cells were cultured and divided into the control group,the model group and the administration group.The control group was cultured with normal hippocampal neurons in the culture medium,The model group was cultured by adding ischemia-reperfusion injury hippocampal neurons to the culture medium.The administration group cultured hippocampal neurons with ischemia-reperfusion injury in the culture medium,and at the same time,1.25 µmol/L edaravone and dexcamphene were added to the interference group.The expression levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA),and the expression levels of TLR4 and nuclear factor-κB(NF-κB)were detected by Western blotting.Results The expression levels of IL-6 and TNF-α in the model group were higher than those in the contro group,and the expression levels of IL-6 and TNF-α in the administration group were lower than those in the model group,the differences were statistically significant(P<0.05).The expression levels of TLR4 and NΚ-κB in the model group were higher than those in the contro group,while the expression levels of TLR4 and NΚ-κB in the administration group were lower than those in the model group,the differences were statistically significant(P<0.05).Conclusion Edaravone and dexcamphene can alleviate the inflammatory response after cerebral ischemia-reperfusion injury and reduce the levels of inflammatory factors by regulating the TLR4/NF-κB signaling pathway.
