葛根芩连汤通过sCD14/TLR4/NF-κB通路调控肠源性内毒素血症肠上皮细胞炎症因子的作用
Gegen Qinlian Decoction regulates inflammatory factors in intestinal epithelial cells with enterotoxemia through sCD14/TLR4/NF-κB pathway

Abstract
Objective To investigate the regulatory effect of Gegen Qinlian Decoction on inflammatory factors in intestinal epithelial cells of intestinal endotoxemia(IETM)through the soluble CD14(sCD14)/Toll like receptor 4(TLR4)/nuclear factor kappa B(NF-κB)signaling pathway.Method In this study,a lipopolysaccharide(LPS)-induced IETM model was established using the human intestinal epithelial cell line SW480.According to the different intervention doses of Gegen Qinlian Decoction,the experiment was divided into blank control group,model control group,low-dose group(10 μg/ml),medium dose group(20 μg/ml),high-dose group(40 μg/ml),and positive drug control group(20 μg/ml berberine).After treatment,the secretion levels of inflammatory cytokines interleukin(IL)-1,IL-6,IL-8,tumor necrosis factor alpha(TNF-α),and interferon gamma(IFN-γ)were detected.Meanwhile,the mRNA and protein expression levels of key molecules such as sCD14,TLR4,and NF-κB were detected by quantitative polymerase chain reaction and Western blotting.Result Except for IL-1 and IL-6,the effect of Gegen Qinlian Decoction on reducing the levels of other inflammatory factors was enhanced with the increase of dose(P<0.05),showing a certain dose dependence.Except IL-1,there was no significant difference in the expression levels of other inflammatory factors between the high dose group and the positive drug control group(P>0.05).Compared with the model control group,Gegen Qinlian Decoction significantly reduced the expressions of sCD14,TLR4,NF-κB and other inflammation-related genes and proteins,also showing a dose-dependent(P<0.05),and there was no statistical difference in the expression of these inflammatory markers in the high-dose group compared with the positive drug control group(P>0.05).Conclusion Gegen Qinlian Decoction has a significant regulatory effect on the inflammatory response of intestinal epithelial cells through the sCD14/TLR4/NF-κB signaling pathway,providing a new strategy for the treatment of intestinal endotoxemia.关键词
