摘要

Abstract

[Objective]The core collection of Pinus sylvestris var.mongolica on sandy lands was constructed on the basis of phenotypic traits and SSR markers,so as to provide foundations for effective management and utilization of its germplasm resources.[Method]Phenotypic traits were investigated in 517 germplasms from 31-year-old plantations of P.sylvestris var.mongolica in Horqin Sandy Land,including the diameter at breast height,tree height,height to live crown base,crown width,individual tree volume,stem form,crown shape and branchiness,and their genetic diversity were assessed using 12 pairs of SSR primers.The core collection were constructed based on these data of phenotypic traits and SSR m,arkers using allele number maximization strategy in conjunction with the principles of extreme phenotype and rare allele prioritization.The representativeness and validity of the core collection were further evaluated through difference tests for parameters of phenotypic variation and genetic diversity as well as analysis of principal coordinate plots.[Result]There are rich phenotypic variation and genetic diversity in collected germplasm resources of P.sylvestris var.mongolica,with the coefficients of variation and the phenotypic diversity index(H′)for each trait ranging from 13.01%to 48.21%and from 0.913 to 2.064,respectively.A total of 97 alleles(Na)are detected at 12 SSR loci,and the mean values of the number of effective alleles(Ne),Shannon-Wiener diversity index(I),observed heterozygosity(Ho),expected heterozygosity(He),and the polymorphic information content(PIC)are 3.324,1.189,0.511,0.556 and 0.512,respectively.A total of 46 core collections are obtained at a sampling rate of 8.90%,and their mean difference percentage(MD),variance difference percentage(VD),coincidence rate of range(CR),and variable rate for the coefficient of variation(VR)are 1.42%,46.33%,100.00%,and 135.34%,respectively.The core collection exhibit significantly higher coefficients of H′,Ne,I,He,and PIC than the initial collection,and the core collections are uniformly distributed throughout the initial collection.The unique DNA fingerprints are also constructed for 46 core germplasm at 11 loci.[Conclusion]The 46 core germplasm collection can effectively represent the phenotypic variation and genetic diversity of the initial collection.
