摘要

Abstract

[Objective]To explore the effects of red light on the growth and lignin content of Populus yunnanensis cuttings,as well as the expression levels of key genes in biosynthetic pathways,through the treatment of red and white light quality,so as to provide a theoretical basis for revealing the molecular mechanism of P.yunnanensis responding to red light.[Method]The cutting seedlings of P.yunnanensis treated with red light and white light for 40 days were used as materials,and the growth indicators,photosynthetic products,lignin,cellulose and hemicellulose contents were determined.The stem segments were collected for transcriptome sequencing,and RT qPCR technology was used to verify the screened differential genes.[Result]The plant height and internode stem length of P.yunnanensis cuttings under red light treatment were higher than those under white light treatment,but the internode stem diameter is lower than those under white light treatment;the thickness of xylem,the contents of total soluble sugar,starch and sucrose,and the contents of cellulose,hemicellulose and lignin under red light treatment are significantly lower than those in white light treatment;A total of 3 002 differential genes obtained by transcriptome sequencing,and the differential genes mainly enriched in the biosynthesis of secondary metabolites,phenylpropanoid biosynthesis,plant hormone signal transduction,starch and sucrose metabolism and other pathways.RT-qPCR results showed that the relative expression of TPSII,MYC2,IRX8,CESA8,SNRK1 and the key genes of lignin synthesis 4CL1 and CCR1 were consistent with the transcriptome data,and the gene expression of 4CL1 and CCR1 under red light treatment is lower than that under white light treatment.[Conclusion]Under red light treatment,the key regulatory genes 4CL1 and CCR1 of lignin synthesis species were down regulated,and the lignin content decreased,which provided a preliminary basis for revealing the growth changes of P.yunnanensis cuttings regulated by red light.
